Omschrijving The gender related diseases such as breast cancer and osteoporosis do have in common that there is imbalance between apoptosis and proliferation. In the case of breast cancer there is an increased proliferation rate and decreased apoptosis resulting in tumor growth, while in contrast in osteoporosis apoptosis in osteoblasts and proliferation in osteoclasts is increased, leading to decreased bone tissue homoeostasis.
In the clinical setting different drugs are already in use for the treatment of the above mentioned gender related diseases, however the single use, combination or sequencing of the different approved medications can only be tested adequately in large randomized clinical trials.
In our laboratory setting we perform measurements of proliferation and apoptosis in three human breast cancer cell lines and human osteoblasts and osteoclasts after the administration of different compounds such as steroidal hormones, aromatase inhibitors and bisphosphonates. These drugs are administered as single compounds or in different combinations. Thereafter the ratios of apoptosis versus proliferation are calculated.
